عنوان فارسی بررسی بیان ژن های CCDC۲۶ و C-Kit در رده های سلولی لوسمی لنفوبلاستیک حاد
چکیده فارسی مقاله زمینه و هدف: لوسمی لنفوبلاستیک حاد، شایع ترین سرطان در کودکان و یکی از عوامل اصلی مرگ و میر آنها در دنیاست. در این مطالعه به بررسی بیان ژن RNA غیر کدکننده ی طویل CCDC26 و همچنین ژن پایین دست آن C-Kit در رده های سلولی لوسمی لنفویلاستیک حاد پرداخته ایم. روش بررسی: این مطالعه از نوع تجربی می باشد. در این مطالعه از رده های سلولی لوسمی لنفوبلاستیک حاد T شامل RPMI 8402  و Jurkat و از رده های سلولی لوسمی لنفوبلاستیک B شامل Daudi  و Ramos استفاده شد. جهت بررسی بیان ژنها از تکنیک qPCR استفاده گردید. تجزیه و تحلیل نتایج با استفاده از آزمون آنوای یک طرفه انجام شد.  یافته ها: در رده های سلولی  RPMI8402 و Ramos افزایش بیان معنادار CCDC26 در مقایسه با بیان ژن C-Kit دیده شد(0/05>p) و در رده سلولی RPMI8402 افزایش بیان ژن CCDC26 نسبت به نمونه ی کنترل، معنادار بود(0/05>p). اما در رده های سلولی Jurkat و Daudi افزایش بیان معنادار ژن C-Kit در مقایسه با بیان ژن CCDC26 و همچنین نمونه کنترل دیده شد(0/05>p). نتیجه گیری: شواهد نشان می دهد که LncRNA نقش مهمی به عنوان یک مارکر در تشخیص و پیش آگهی برخی سرطان ها دارند. یافته های فوق نشان می دهد که ژن CCDC26 که در بالادست ژن C-Kit است، احتمالاً نقش تنظیم کنندگی برای این ژن دارد. ژن C-Kit در رشد و تکثیر سلول های خونی نقش دارد.

چکیده انگلیسی مقاله Background and Aim: Acute Lymphoblastic Leukemia is the most common cancer in children and is one of the main causes of their mortality. In this study, we examined the expression of long noncoding RNA CCDC26 gene and its downstream C-Kit gene in acute lymphocytic Leukemia cell line. Materials and Methods: This is an experimental study. In this study, the acute Lymphoblastic Leukemia cell lines T (RPMI 8402 and Jurkat) and the cell lines of Lymphoblastic B Leukemia (Daudi and Ramos) were used. After culture, RNA extraction and cDNA synthesis were performed. In order to study the expression of CCDC26 and C-Kit genes, Real Time PCR technique was done. One-way ANOVA test was performed to analyze the results. Results: In RPMI8402 and Ramos cells, a significant increase in expression of CCDC26 was observed in comparison with the expression of the C-Kit gene (P< 0.05) and in the RPMI8402 cell line, the expression of the expression of the CCDC26 gene was significant (p< 0.05) But in the Jurkat and Daudi cell lines, the significant increase of C-Kit gene expression was observed as compared to the expression of the CCDC26 gene as well as the control sample (pv< 0.05). Conclusions: Evidence suggests that lncRNA plays an important role as a marker in the diagnosis and prognosis of some cancers. The findings suggest that the CCDC26 gene located on the upstream of the C-Kit gene has a regulating role for this gene. The C-Kit gene plays a role in the growth and proliferation of blood cells.
