چکیده انگلیسی مقاله Background: In the past decades many researches have been conducted to comprehend anti-cancer effects of curcumin. Although anti proliferative properties of curcumin on cancerous cells is revealed, the clinical application of this gold substrate is limited yet. This obstruction is mostly due to low solubility, inefficient bioavailability, rapid metabolism and improper uptake. In this study, a novel biodegradable gemini surfactant (Gs) was synthesized and curcumin molecules were encapsulated within polymer in order to overcome physicochemical limitations of curcumin.Methods: gemini surfactant-curcumin (Gs-Cu) nanoparticles were prepared using nano precipitation method. Size and poly dispersity index of nanoparticles were determined with Dynamic Light Scattering (DLS) technique. The release profile of curcumin from polymer matrix was studied. MTT assay and cellular uptake of Gs-Cu on MDA-MB-231 cells were investigated in vitro.Results: The Gs polymer was capable of forming polymersomes in aqueous solution and the narrow size distribution was obtained (PDI≅0.3). Encapsulation efficiency was calculated about 87%. As a result of incorporation of curcumin in polymer matrix, sustained release profile was observed. Moreover, Gs-Cu complex showed more cytotoxicity in compared to free curcumin because of higher rate of cellular uptake.Conclusions: The data indicate that Gs polymersomes can be regarded as nano carrier for hydrophobic curcumin molecules.
