عنوان فارسی بررسی نیمه کمی بیان ژنKAI۱ در مراحل مختلف mRNA بافتی بیماران ایرانی مبتلا به سرطان بافت سنگفرشی مری و ارتباط آن با فاکتورهای دموگرافی و پاتولوژی بیماران
چکیده فارسی مقاله پیش‌زمینه و هدف: ژن KAI1 یک ژن شناخته‌شده سرکوبگر متاستاز است که با مهار تحرک و تهاجم سلول‌های سرطانی اولیه، متاستاز را سرکوب می‌کند. ارزیابی بیان KAI1 در طیف وسیعی از تومورها، نشان داده کهKAI1 در همه بافت‌های نرمال بیان دارد اما میزان بیان mRNA آن در بافت‌ها یکسان نیست. هدف از انجام این مطالعه، ارزیابی نیمه کمی بیان ژنKAI1 در mRNA بافتی بیماران ایرانی مبتلا به سرطان بافت سنگفرشی مری و ارتباط خاموش یا روشن بودن این ژن در هر یک از مراحل سرطانی مری و سایر فاکتورهای دموگرافی و پاتولوژی بیماران به‌منظور تعیین یک مارکر مفید در شناسایی مراحل توموری به‌ویژه متاستاز و تهاجم در این بیماران بود. مواد و روش کار: بدین منظور 35 نمونه توموری تازه و 12 نمونه نرمال از 35 بیمار مراجعه‌کننده به بیمارستان امام خمینی تهیه شد و آزمایش RT-PCR بر روی cDNA سنتز شده حاصل از mRNA بافت‌ها با پرایمرهای بتااکتین و ژنKAI1 انجام شد. یافته‌ها: نتایج نشان داد در 69.2درصد نمونه‌ها بیان ژنKAI1 روشن و در 30.8درصد نمونه‌ها بیان این ژن خاموش بود. به‌طور جزئی‌تر نمونه‌های مرحله 3 و4 و نمونه‌های توموری واجد متاستاز اصلاً بیانی برای این ژن را نشان نداد. تجزیه‌وتحلیل آماری مقایسه بیان ژنKAI1 در چهار گروه نرمال، مرحله یک، دو و سه توموری نشان داد که اختلاف معنی‌داری در بیان ژنKAI1 در این گروه‌ها وجود دارد (P< 0.05). نتیجه‌گیری: ارتباط معناداری بین بیان نیمه کمی ژنKAI1 با مرحله توموری نمونه‌ها(P< 0.05) و با متاستاز(P=0.00015) مشاهده شد.

عنوان انگلیسی SEMI-QUANTITATIVE EVALUATION OF KAI1 GENE EXPRESSION AND CORRELATION BETWEEN STAGES OF TUMORS, DEMOGRAPHIC AND PATHOLOGIC FACTORS OF PATIENTS WITH SQUAMOUS CELL CARCINOMA OF ESOPHAGUS
چکیده انگلیسی مقاله  Background & Aims: KAI1 is a tumor suppressor gene and inhibitor of metastasis in a wide range of malignancies. While it is ubiquitously expressed in normal tissues, KAI1 expression is down regulated in tumors. The present research proposal aims semi quantitative evaluation of KAI1 mRNA expression in Iranian patients with Squamous Cell Carcinoma of Esophagus(SCCE) on finding a correlation between expression levels of KAI1 and stage of tumorigenesis, especially metastatasis and invasion of SCCE as well as demographic and pathologic factors of patients. Materials & Methods: In this study, KAI1 and β-Actin mRNA levels was detected by RT-PCR on 35 tumor tissues and 12 normal tissues as fresh samples obtained from 35 patients referring to Imam Khomeini Hospital. Results: Accordingly, 69.2% and 30.8% of the sample were on and off for KAI1 gene expression, respectively. The samples in stages 3, 4, and metastatic phases showed no expression of this gene. Statistical analysis showed that there was a significant differences of the KAI1 expression between four groups of samples normal, stage 1, 2, 3 (P < 0.05). Conclusion: There is a significant difference between semi-quantitative of KAI1 expression and stages of tumor samples (P < 0.05) as well as semi-quantitative of KAI1 expression and metastasis (P = 0.00015). The outcome of the present research proposal would be applicable as a suitable mean for clinicians in order to better treat Esophageal Squamous-Cell Carcinoma. SOURCE: URMIA MED J 2015: 25(12): 1127 ISSN: 1027-3727
