عنوان فارسی نقش کاهش فعالیت بدنی به شکل درد نروپاتیک در افزایش بیان فاکتور رشد عصبی در عصب سیاتیک
چکیده فارسی مقاله مقدمه: درد نوروپاتیک با تاثیر عمیق بر کیفیت زندگی و عملکرد روزانه بیماران همراه بوده و هزینه‌های درمانی بالایی را اعمال می‌کند. در این بیماری به‌خاطر طبیعت تخریبی اعصاب درآن، توجه بالایی به نوروتروفین‌ها به‌خاطر تاثیرات احتمالی آن­ها بر حیات، رشد، توسعه و تنظیم عملکرد نورون‌ها به‌عنوان یک راه درمانی احتمالی شده است. بنابرین هدف از انجام پژوهش حاضر بررسی اثرات مزمن کاهش فعالیت بدنی به شکل درد نوروپاتیک در بیان ژن NFG (Nerve Growth Factor) در عصب سیاتیک رت‌های دچار لیگاسیون عصب نخاعی است. روش‌ها:‌ 10 سر موش صحرایی نر نژاد ویستار با میانگین وزن 30±250 گرم به 2 گروه کنترل سالم (5n=) و گروه درد نوروپاتیک (SNL) (5n=) تقسیم شدند. طی شش هفته، آزمون‌های رفتاری درد نوروپاتیک در گروه‌های پژوهشی به‌طور مستمر انجام شد. در پایان 6 هفته تغییرات بیان ژنNGF در عصب سیاتیک با تکنیک Real time PCR اندازه‌گیری و با روش2-&Delta&DeltaCT محاسبه شد. یافته‌ها: پس از 6 هفته، آزمون­های رفتاری درد نوروپاتیک آلوداینیای مکانیکی و پردردی حرارتی نشان داد که در گروه لیگاتور بندی آستانه تحریک درد نسبت به گروه کنترل به‌طور معنی‌داری کمتر بود (05/0&le P). همچنین میزان بیان ژن NGF در عصب سیاتیک در گروه لیگاتور بندی شده به‌طور معنی‌داری نسبت به گروه کنترل افزایش نشان داد (05/0&le P). نتیجه‌گیری: یافته‌های مطالعه حاضر نشان می‌دهند که توسعه درد نوروپاتی و کاهش فعالیت بدنی در اثر آن با افزایش درون‌زاد NGF همراه است. هرچند در مطالعه حاضر مشخص نیست افزایش در بیانNGF از فعالیت کاهش یافته حاصل از درد نوروپاتی است یا از ذات آسیب عصبی، با این حال مسلم است افزایش بیان NGF، با درد نوروپاتیک همراه است.

عنوان انگلیسی THE EFFECT OF DECREASED ACTIVITY IN NEUROPATHIC PAIN FORM ON INCREASED NERVE GROWTH FACTOR GENE EXPRESSION IN SCIATIC NERVE
چکیده انگلیسی مقاله Background: Neuropathic pain is associated with a profound impact on quality of life and daily activity in patients and caused high medical costs. Because of the nerves destructive nature in this disease, the neurotrophins are high regarded for their possible effects on survival, growth and development and neuronal functions as a possible therapeutic strategy. So the aim of this study is investigation of the chronic effects of decreased activity in neuropathic pain form on the NFG gene expression in the sciatic nerve of Spinal nerve ligated rats. Methods: Ten adult male Wistar rats in the weight range of 250±30 gr randomly were divided into two groups including healthy control (C), ligation sciatic nerve ligation. Over the six weeks neuropathic pain behavior tests were conducted continually in groups. In the end change of NGF gene expression in sciatic nerve was measured with Real time PCR technique and calculated using the 2-&Delta&DeltaCT method. Results: After 6 weeks, neuropathic pain behavior tests showed that pain threshold of thermal hyperalgesia and mechanical allodynia in the SNL group was significantly lower than the control group (P&le0.05). In addition, NGF gene expression in sciatic nerve ligation group compared to controls increased significantly (P&le0.05). Conclusion: The findings of this study suggest that the development of neuropathic pain and decreased physical activity is associated with increased endogenous NGF. Although it is not clear that increase in NGF expression is due to the nature of neuropathic pain or nerve damage, it is clear that excessive expression of NGF is associated with neuropathic pain.
