عنوان فارسی تاثیر ۶ هفته مکمل سازی کوئرستین بر بیان برخی شاخص‌ های آنژیوژنزی بافت تومور در موش‌ های ماده مبتلا به سرطان پستان
چکیده فارسی مقاله مقدمه: کند کردن فرآیند آنژیوژنز در بافت تومور، در روش ‏های جدید درمان سرطان پستان مورد توجه است. هدف این پژوهش، بررسی تاثیر مکمل کوئرستین بر بیان عوامل کلیدی آنژیوژنز در بافت تومور موش‏ های مبتلا به سرطان پستان است. روش بررسی: به 16 سر موش بالب سی ماده، سلول‏ های سرطانی وابسته به گیرنده استروژن MC4L2 زیر جلدی تزریق شد و سپس به دو گروه تومور (T) و تومور+کوئرستین (TQ) تقسیم شدند. موش‏های گروه TQ، 6 هفته، 3 روز در هفته، 110 میلی‏ گرم/کیلوگرم محلول کوئرستین زیر صفاقی تزریق شد. 48 ساعت پس از آخرین تزریق، موش‏ ها کشته شدند و بافت تومور آن‏ها برداشته و در نیتروژن مایع فریز شد. و در دمای 70- درجه نگهداری شدند. آنگاه بیان ژن‏ های ANGPT-1، ANGPT-2، TIE-2 و VEGF-A به روش REAL TIME-PCR سنجیده شد.ΔCt ، ΔΔCt، Fold chang و آزمون من ویتنی- در سطح معناداری)P< 0/05(- با نرم افزار GENEX محاسبه شد. یافته‏ ها: نتایج نشان می‏ دهد مکمل سازی کوئرستین باعث کاهش معنادار بیان ژن‏ های  (P< 0/00)،TIE-2 (P< 0/00) و VEGF-A (P< 0/00) درگروه TQ مقایسه با گروه T شده است. اما بیان ANGPT-1 به رغم افزایش، معنادار نبود. نتیجه‏ گیری: مکمل سازی کوئرستین احتمالا باعث مهار آنژیوژنز تومور و در نتیجه رشد تومور پستان می‏ شود.

عنوان انگلیسی Effect of 6 Weeks of Quercetin Supplementation on Expression of Some Tumor Angiogenesis Markers in Female Mice with Breast Cancer
چکیده انگلیسی مقاله Introduction: Slowing down the process of angiogenesis in the tumor tissue is an emerging area in the treatment of breast cancer. The purpose of this study was to investigate the effect of quercetin supplementation on the expression of key angiogenesis factors in the tumor tissue of mice with breast cancer. Methods: Sixteen female BALB /c mice were injected subcutaneously with estrogen receptor dependent breast cancer cells MC4L2 and divided into two groups of tumor (T) and tumor + quercetin (TQ). The mice in the TQ group were injected subcutaneously with 110 mg/kg quercetin solution for 6 weeks, 3 days a week. Forty-eight hours after the last injection, the animals were killed and their tumors were removed, frozen in liquid nitrogen and stored at -70° C. ΔCt, ΔΔCt, fold change and Mann-Whitney test at the significance level of (P< 0.05) were calculated with GENEX software. Results: The results showed that quercetin supplementation significantly decreased the expression of ANGPT-2 (P< 0.00), TIE-2 (P< 0.00) and VEGF-A (P< 0.00) genes in the TQ group compared with the T group. But ANGPT-1 expression increased, although this increase was not significant. Conclusion: Quercetin supplementation may inhibit tumor angiogenesis and breast tumor growth.
