عنوان فارسی ارزیابی روش‌های مختلف استخراج DNA از ماهیان قزل‌آلای‌رنگین‌کمان طلایی پرورشی (Golden Rainbow Trout: Oncorhynchus mykiss) به منظور مطالعه مولکولی
چکیده فارسی مقاله استخراج DNA یک گام معمول در بسیاری از مطالعات بیولوژیکی است و روش­های مختلفی جهت جداسازی مولکول DNA از مواد بیولوژیکی ابداع شده است. بسیاری از این روش­ها دارای سه هدف: 1- لیز سلول و خروج اسید­نوکلئیک از مایع درون سلولی، 2- از بین بردن مولکول­های آلی و معدنی غیر از اسید­نوکلئیک در مایع درون سلولی و 3- به حداقل رساندن تخریب اسید­نوکلئیک­ها در طی مراحل خالص­سازی می­باشند. ماهی قزل­آلای­رنگین­کمان از جمله گونه­های اهلی شده آزاد ماهیان بوده و امروزه مهم­ترین گونه ماهی سردآبی در کشور است. جهت انجام این پژوهش 4 روش استخراج: 1- کیت استخراج شرکت Gena All،  2- Trizol Reagent، 3- روش سنتی حرارت و آنزیم پروتئیناز­K و 4- فنل­کلروفرم مورد بررسی قرار گرفت و مشخص شد که بهترین روش استخراج از لحاظ کیفیت و کمیت DNA استخراج شده توسط دستگاه نانودراپ و ژل آگاروز 1 در­صد و همچنین از لحاظ کارآمد بودن PCR مربوط به روش کیت استخراج DNA شرکت Gene All کشور کره جنوبی می­باشد.

چکیده انگلیسی مقاله DNA extraction is a routine step in many biological studies and variety of methods have been established to isolate DNA molecules from biological materials. most methods share three aims: 1- the comprehensive lysis of cells and extraction of intracellular nucleic acids into aqueous solution, 2-  the removal of non-nucleic acid organic and inorganic molecules from resultant aqueous extracts  and 3- the minimization of nucleic acid losses throughout this purification process. Rainbow trout fish, including salmonid species have been domesticated and today the most important cold-water fish species in the country. To accomplish this study 4 Extraction 1. Extraction Kit Company Gena All 2- Trizol Reagent 3- traditional method of heating and Proteinase K 4- phenol chloroform were studied  and It was found that the The best extraction method in terms of quality and the quantity of DNA extracted by 1 percent agarose gel and Nanodrop and Also in terms of the effectiveness of PCR for DNA extraction kit method is the enterprise Gene All Country South Korea.
