عنوان انگلیسی Evaluation of different decalcifying agents on histochemical and immunohistochemical staining properties of canine osseous tissue
چکیده انگلیسی مقاله Introduction: In routine histopathology, decalcification of calcified tissues is a principal step before tissue processing. Considerable studies have been made to compare the decalcifying substances in order to meet the criteria including complete removal of calcium, minimal damage to tissues, non-impairment to subsequent staining, and working at a reasonable speed. Thus, the purpose of this study is to evaluate the effect of several decalcifying agents on the morphological and antigenicity preservation in canine mandibular tissue.
Methods: Four different decalcifying solutions, namely 5% nitric acid (NA), 10% and 20% formic acid (FA) plus 10% EDTA were employed to remove mineral salts from specimens harvested from mandibular osseous tissue of four healthy Mongrel dogs (32 samples). They were compared in respect to the time period of decalcification, ease of tissue slicing, morphological organization after alizarin red S staining and osteocalcin antigenicity preservation.
Results: With regard to the short period of decalcification and ease of sectioning, 5% NA gives better results than 10% and 20% FA, and 10% EDTA (pH 7.4). In the case of morphology preservation and antigenicity maintenance of the tissue samples, 10% EDTA is found to be the most optimal solution followed by 10% and 20% FA, and 5% NA.
Conclusion: Our findings support the use of EDTA as the highly-preferred choice for decalcification of canine osseous tissue if IHC is aimed despite of its time-consuming trait. However, FA 20% and NA 5% are the choice for general histological staining procedures.
